Abstrakt česky: Tento přehledový článek popisuje přípravu derivátů kyseliny betulinové modifikovaných na A-kruhu, jejich biologickou aktivitu, komentuje SAR a mechanizmus účinku. Měl by sloužit pro orientaci v dané problematice.
Abstrakt anglicky: This focused review summarizes recent advances in the synthesis of Betulinic acid derivatives with modified A‐ring. It also focuses on their development as potential therapeutics for treatment of cancer and other diseases. Betulinic acid (BA) has therapeutically interesting biological activities such as anti‐tumor, anti‐HIV, anti‐protozoal, and hepato‐ or nephro‐protective, however, its clinical use is severely limited by higher IC50 and worse pharmacological parameters then those in currently used drugs. On the other hand, semi‐synthetic analogues of BA surpassed their parent and are much more suitable for the potential clinical use. Among them, compounds with modified A‐ring are the most promising. This review should serve the other researchers as a guide to the synthesis of new BA derivatives with much higher chance of being developed as a drug than BA. The general audience may find this review interesting because we describe the structural motifs responsible for the bioactivities.
