Abstrakt anglicky: Recently, click chemistry has been used extensively to obtain a diverse range of functionalized nucleic acids and oligodeoxynucleotides. The modification of nucleosides and oligodeoxynucleotides has been performed on both the monomer level and as a post-oligodeoxyribonucleotide synthesis modification. It has recently been found that one major contributor, if not the primary, to the overall duplex stability of both DNA and RNA is the nucleobase π-π-stacking. In light of these findings we present work on introduction of aromatic moieties in the major groove of the DNA duplex to obtain increased overall duplex stability by site-specific placement of these functionalities. We therefore anticipate that heavily modified DNA duplexes of this kind may find use in nanobiotechnology. From our initial work with modified DNA duplexes each having four consecutive modifications, we observe an increase in overall duplex stability of +5 °C pr. modification.
